Major improvements in instrumentation and specimen preparation have brought SEM to the fore as a biological imaging technique. In FESEM, a field-emission cathode placed in the electron gun of a scanning electron microscope provides narrower probing beams and high electron energy. The result is improved spatial resolution and minimized sample charging and damage. Images produced are less destroyed and have a spatial resolution down to 1.5 nm, three to six times better than conventional SEM.
Although this imaging technique has undergone tremendous developments, it is still poorly...

All sexually reproducing organisms produce primordial germ cells, a small population of cells that differentiate into gametes of either sex and carry to- potency, an ability to develop into an entire new organism. The study of germ cells has undergone enormous advances in recent years and has entered into an explosive phase of new discoveries with the introduction of transgenic te- nologies and nuclear cloning. Basic knowledge and techniques developed for lower vertebrate and invertebrate systems have facilitated the study of higher vertebrates, including humans. Many experiments that have...

Basic knowledge and te- niques developed for lower vertebrate and invertebrate systems have facilitated the study of higher vertebrates, including humans. Many experiments that have first been performed on lower vertebrates provided the tools and strategies that could later be applied to other less readily available mammalian systems. The discovery of centrosomes in ascidians and sea urchin eggs...

Salmonella: Methods and Protocols presents detailed methods on a variety of aspects of Salmonella research, focusing on those which provide landmarks for future discovery. Highlighted topics include molecular genotyping, rapid and accurate new methods for sensitive determination of antibiotic resistance profiles, the characterization of Salmonella-specific cell antigens, and Salmonella-host cell interactions. Detailed methods for studying Salmonella motility are also presented. Each chapter provides a short overview of the topic, which is followed by detailed explanations of techniques...
