The present study deals with the determination of the effects of source geometry, gamma source activity and source volume on the intrinsic uniformity. A comparison of intrinsic uniformity for different methods for preparation of point source with extrinsic uniformity was done. Statistics was applied to analyze the experimental data. Different methods were used to make best optimized spherical point source for measurement of intrinsic uniformity. For this purpose I was used cotton swab point source and syringe point source for best geometry and performed the test at 5% UFOV recommended by NEMA (1994), Elkamhawy et al (2000) and Zanzonico (2008) and also according to the technique recommended by siemens (2002) and AAPM (1987). And conclude the following results Syringe method produced a uniform flood source and the geometry of the point source retained and best results of intrinsic uniformity. Extrinsic uniformity was time deserving due to its low activity and it was not preferable for the system uniformity detection. Intrinsic uniformity results at 5 % UFOV which is recommended by NEMA (1994) and Zanzonico (2008) is strongly preferable.
