Extremophiles are microorganisms that can thrive in most extreme environments and constitute a potential source of proteins that are stable under such conditions. Proteins produced by these organisms have evolved specific mechanisms for their solubility at high salt concentrations. It will be interesting to study whether compatible solutes can replace high salt concentration in haloarcheal system without affecting protein function and stability. HS4 gene was already cloned in to pTZ57R/T. Thus, Sub cloning of HS4 gene from pTZ57R/T clone into pET28a (+) expression vector done successfully for protein expression. Overexpression studies of protein was done. However, high level production of desired protein could not be achieved. A complete understanding of the mechanism may revolutionize the use of these novel proteins in research and industrial processes.
