Objective: The present study was undertaken to evaluate the anti-stress effect of RNH-12 on experimental induced stress in rat.Methodology: The anti-stress effect was evaluated on cold restraint induced stress, and swimming endurance stress induced. RNH-12 was administered orally at two different doses of 40 and 60 mg/kg. Withania somnifera, a known anti-stress formulation (500 mg/kg p.o) was used as the standard drug. Results: Both the doses of RNH-12 showed good anti-stress effect in all the tested animal models. The RNH-12 reduced the development of cold restraint induced gastric ulcers and prevented the biochemical changes induced by swimming endurance and swimming endurance stress such as increase in plasma levels of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, glucose, cholesterol and Cortiosol. RNH-12 was also effective in increasing the latency of anoxic stress induced convulsions in rat. Conclusion: The investigational drug of RNH-12 at doses of 40 and 60 mg/kg orally possesses good anti-stress activity.
