Tissue culture has been subject of many studies in recent decades especially for plants that their growth and propagation is not easy. Sumac belongs to genus of Rhus (Anacardiaceae). Vitality of Sumac seed is very low and it naturally propagates through sprig. In this research the best media for in vitro regeneration and rooting of Sumac has been studied. Buds as explants were sterilized and transferred to callus induction media. Callus induction was initiated on MS basal media supplemented with IAA, BA, BAP, TDZ, IBA. Results indicated that the most callus induction and regeneration was on MS fortified with 1 mg/l BA + 0.5 mg/l IAA and the least was 1.5 mg/l BA + 0.1 mg/l IAA that were 60% and 0 respectively. Half , quarter and full strength MS medium supplemented with IAA, NAA, IBA were used for rooting. The results showed significant differences between rooting treatment. The most rooting observed on full strength MS medium supplemented with 1 mg/l IBA and the least observed on half strength MS supplemented with 8 mg/l IBA that were 44.5% and 2.66% respectively. The best treatment for transformation to rooting was 1 mg/l BA + 0.5 mg/l IAA.
