Gladiolus Cultivar Friendship is highly susceptibleto soil born Fungus Fusarium oxysporum. Highlyresistant cell-lines were regenerated throughin-vitro selection at callus and cell suspensionlevel. Cell suspensions of all four Gladioluscultivars were found highly sensitive to fusaricacid. Gradual increase in fusaric acid concentrationsto the cell-suspension cultures decreased cell growthconsiderably. The cormlets of all in-vitro selectedcell lines of "Friendship" were inoculated with aconidial suspension of the F. oxysporum. The fourselected cell lines showed the same response whetheror not inoculated with conidia of the F.oxysporum. Plantlets of all of the selected celllines exhibited significant growth as compared withcontrol. RAPD markers were used to estimategenetic diversity among the selected Fusariumresistant cell-lines. Marker data was generatedthrough three successive generations. The number offragments per primer ranged from 4 to 10. The totalsize of the amplified products varied from 200bp to1800bp. All the primers were found to be polymorphicand produced upto 62% polymorphism.
