ARTERIAL WALL LIPOGENESIS: REGULATION OF ITS EXPRESSION AND ANOMALIES IN INSULIN RESISTANCE AND DIABETES We studied the expression and regulation of lipogenesis in aortas and CMLV and determined whether it is altered by insulin resistance and diabetes. Obese Zucker (ZO) rats, diabetic rats, and Psammomys obesus rats accumulate more lipids in their aortas than their controls. However, the expression of lipogenesis genes and those involved in fatty acid uptake is not increased. A similar result was found in human endarterectomy specimens. The adipogenic environment, glucose, or insulin alone moderately stimulate lipogenesis only in control Zucker CMLs; no effect was observed in ZO CMLs. We have shown that the effects of TO901317 on lipogenesis in CMLV are due solely to the activation of the nuclear receptor LXR ; PXR has no effect. In conclusion, lipogenesis is not increased in the arterial wall during insulin resistance and diabetes.
