Spectrophotometric method was developed for the determination of BetaxololHCl in STF at 222.5 nm. FTIR spectrum of pure drug and drug-polymer mixture revealed no chemical interaction. Total fifteen batches of in-situ gelling system using gelrite and sodium alginate- HPMC E50LV for ion activated mechanism were prepared and passed through nylon membrane filter and terminally were sterilized by autoclaving at 121°C and15lb pressure for 20 min. Various in vitro parameters like gel retention time, clarity, pH, drug content, in vitro drug diffusion, test for sterility, viscosity determination were evaluated. Stability study of G6 and SA6 formulations were performed and G6 not showed any significant changes in all parameters evaluated while SA6 (alginate base) batch was developed haziness so it was discarded after the one month of stability studies and so, G6 batch was selected as the best formulation. Ocular irritation studies were evaluated for the best formulation selected on the basis of in vitro parameters and stability studies conducted.
